UMD TP53 Mutation Database

The 2008_R2 release of the UMD_TP53 Mutation database (2356 references, 27 900 mutations) is now available (November 2008).

Thierry Soussi, project leader
Linn Hjortsberg, database manager
Christophe Béroud, Dalil Hamroun and Jean Michel Rubio-Nevado, Developer

The UMD TP53 database was first released in 1992 (Caron de Fromentel C, Soussi T (1992) TP53 Tumor suppressor gene: a model for investigating human mutagenesis. Genes Chrom Cancer 4: 1-15.)

Since this initial release (300 mutations), the UMD TP53 database has progressed and the second 2008 release contains about 28 000 mutations.

What's new in the 2008 UMD Website site:

  • A better curation of the UMD p53 mutation database to remove artefactual data
  • A novel section devoted exclusively to the p53 status in human cancer cell lines
  • A novel section devoted to the controversy about p53 mutations in stromal cells in breast cancer patients
p53_database

Every p53 mutations database available through the web include publications with artefactual data that will change the outcome of all types of studies.
Using a sophisticated statistical methodology, these publications have been identified (Soussi et al, Clin. Cancer Res., 2006, Soussi et al., unpublished).

This release of the UMD database comes in two versions

i) The uncurated database with all publications
ii) a curated database that has been purged from artefactual data

We strongly advocate against the use of an uncurated database for any analysis.
More information about controversial p53 mutations in breast cancer can be found here

(see this page to understand how and why the p53 database has been curated)

A complete analysis of the p53 mutation database for the most frequent cancers is available ->Link

A novel excel® matrix to check p53 mutations before publications has been developed and can be downloaded -> Link

The UMD p53 Web site is also active for online analysis of every p53 mutant

  1. Biochemical, structural and functional properties of every p53 mutant -> Link
  2. Check the validity of a newly found p53 mutation based on its activity -> Link

You can also look at the nomenclature for p53 mutations at hgvs -> Link
